In the last few years several publications have changed our perception of the structure and function of the TCR/CD3 complex. We review here its stoichiometry, valence, topology and conformational variability, together with the conformational changes that take place after ligand engagement, and the specific role of each CD3 chain in these features. Despite its surprising flexibility, the TCR/CD3 complex endows T cells with an exquisite specificity.
